Can you play two poker sites at once with PT4? If so, how?

Yes you can. It doesn't require any additional steps other than making sure that each site is configured correctly to import hands:

So how do you choose Configure Sites when playing two platforms? One has to be selected. Can you select two?

You don't have to choose. As long as each site is configured correctly via 'Configure -> Sites & Import Options -> Site Configuration' then when you click 'Play Poker -> Get Hands While Playing' the auto-importing will check each poker site folder for hands even if you are playing on more than one site.
